In Europe, 25% of inhabitants suffer from gut-associated diseases. It often starts with the 'Closed Gut' phenomenon. Practically, this means malabsorption and an 85% inactive enteric nervous system.
Furthermore, a Closed Gut creates an excess of noxae (ammonia derivatives), which cannot be adequately detoxified and cause shifts within the various biogenic amines. These ammonia derivatives occur under physiological conditions in animal organisms and have multiple functions; they act as neurotransmitters, co-factors or hormones.
In a Closed Gut, an increased toxin load causes a thickening of the connective tissue to protect the body. Figuratively speaking, the sluice gates close. This means that the therapeutic approach must be started differently than with Leaky Gut Syndrome.
The symptom pattern for a Closed Gut and Leaky Gut are similar, and therefore, based on symptom registration, the wrong therapeutic route can be chosen. For example, mucous membrane-strengthening measures (e.g., with glutamine) are counterproductive in a Closed Gut and often worsen the patient's symptoms. The diagnostics of the Closed Gut/ Leaky Gut screening and mucosal immunity provide clarity about which therapeutic route is necessary.
If you have opted for a Basic Gut Screening, the separate zonulin test provides insight into whether there is a Closed Gut or Leaky Gut.
The underlying condition or the clinical significance of this test is insufficiently supported within mainstream Dutch medicine.
Rationale for this test
Het concept 'Closed Gut' is geen erkend ziektebeeld binnen de Nederlandse reguliere geneeskunde en komt niet voor in NHG-standaarden of FMS-richtlijnen. De beschreven pathofysiologie (verdikking connectief weefsel door ammoniakderivaten, 85% inactief enteraal zenuwstelsel) heeft geen wetenschappelijke basis in de reguliere geneeskunde. Hoewel enkele individuele markers zoals calprotectine en alfa-1-antitrypsine wel klinisch gevalideerd zijn voor specifieke darmaandoeningen, is de combinatie en interpretatie binnen het 'Closed Gut versus Leaky Gut'-framework niet evidence-based.
Do not interpret a result from this test as a diagnosis, and always discuss with your GP which regular diagnostics are appropriate for your symptoms.

Practical information
After ordering we ship the kit to your home. You collect a small sample with the included scoop, keep it cool and return it in the envelope. You receive the result digitally by email.
Lay plastic film over the toilet or use a clean container. Scoop from the middle of the stool and avoid contact with urine or toilet water.
A small scoop with the included spoon is enough — about half a teaspoon. More is not better.
Maximum 24 to 48 hours in the fridge. Send as soon as possible and preferably early in the week, not just before the weekend.
Some analyses (like calprotectin) ask you to avoid NSAIDs or certain supplements 24-48 hours before collection. Check the instructions with your kit for the exact preparation for your test.
Stool analyses typically take about a week after the lab receives your sample. You receive your laboratory report securely by email. It lists the measured values, units, reference ranges and any flags added by the laboratory.
Use the supplied return envelope and place it in the letter-mail bag inside a PostNL service point yourself as soon as possible. Do not hand it to counter staff or request track & trace. The instructions supplied with your test state when it must be sent.
